KissPeptin-10
Also known as KP-10
The shortest fully active fragment of kisspeptin, a hypothalamic peptide that drives gonadotropin-releasing hormone (GnRH) secretion. It is studied as a tool for probing and stimulating the human reproductive axis.

Overview
Kisspeptins are peptides encoded by the KISS1 gene, first identified as a melanoma metastasis suppressor. In 2001 they were shown to be natural ligands of the orphan receptor GPR54, now called KISS1R, and in 2003 loss-of-function GPR54 mutations were linked to hypogonadotropic hypogonadism, establishing kisspeptin signaling as essential for puberty and fertility. Kisspeptin-10 is the C-terminal decapeptide and the minimal sequence with full intrinsic bioactivity. Human studies have used it to stimulate LH release and study GnRH pulsatility, alongside the longer kisspeptin-54. It is not approved as a medicine.
Class: KISS1R (GPR54) agonist peptide
Mechanism of action
How the compound acts, as described in the literature.
Binds KISS1R (GPR54), a Gq-coupled receptor; activation stimulates PIP2 hydrolysis, calcium mobilization, and ERK1/2 and p38 MAP kinase phosphorylation (Kotani 2001).
KISS1R on hypothalamic GnRH neurons drives GnRH release, which in turn stimulates pituitary LH and FSH secretion and downstream sex-steroid production.
In healthy men, kisspeptin-10 raised LH within 30 minutes, and continuous infusion increased LH pulse frequency, pulse size, and serum testosterone (George 2011).
Responses differ by sex and cycle phase: kisspeptin-10 raised gonadotropins in men and in preovulatory women but not in women during the follicular phase (Jayasena 2011).

2003
Human genetics and mouse study
PMID 14573733The GPR54 gene as a regulator of puberty.
Seminara SB, et al. · N Engl J Med
Patients with idiopathic hypogonadotropic hypogonadism carried loss-of-function GPR54 mutations, and Gpr54-deficient mice showed hypogonadotropic hypogonadism, identifying GPR54 as a key regulator of puberty.
2011
Human clinical study
PMID 21632807Kisspeptin-10 is a potent stimulator of LH and increases pulse frequency in men.
George JT, et al. · J Clin Endocrinol Metab
In healthy men, kisspeptin-10 produced a rapid, dose-dependent rise in LH, and continuous infusion increased testosterone, LH pulse frequency, and secretory burst mass.
2011
Human clinical study
PMID 21976724The effects of kisspeptin-10 on reproductive hormone release show sexual dimorphism in humans.
Jayasena CN, et al. · J Clin Endocrinol Metab
Kisspeptin-10 raised LH and FSH in healthy men and in women during the preovulatory phase, but did not alter gonadotropins in women during the follicular phase, showing sexual dimorphism in response.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Human studies are small physiological studies, often with four to six participants per group, and short in duration. Responses depend on sex and menstrual-cycle phase, and in men the LH response declined at the highest level tested. Kisspeptin-10 is investigational and not approved for any indication.
