Snap-8
Also known as Acetyl octapeptide-3, Ac-EEMQRRAD-NH2
Snap-8 (acetyl octapeptide-3) is a synthetic eight-amino-acid peptide modeled on the N-terminal region of the neuronal protein SNAP-25. It is studied in cosmetic research on expression wrinkles.

Overview
Snap-8 is an elongated analog of acetyl hexapeptide-3 (Argireline, Ac-EEMQRR-NH2), with two additional residues: Ac-EEMQRRAD-NH2. Argireline was rationally designed to mimic part of SNAP-25 and interfere with the SNARE complex that drives neurotransmitter release, an approach intended to reproduce, at much lower potency, one effect of botulinum toxin. Snap-8 was developed by the cosmetic ingredient industry along the same lines. Peer-reviewed data on Snap-8 alone are scarce; indexed human studies test it only within multi-ingredient formulations, so its individual contribution cannot be isolated.
Class: SNARE-modulating cosmetic peptide
Mechanism of action
How the compound acts, as described in the literature.
Designed to compete with SNAP-25 for a position in the SNARE complex, the protein assembly required for calcium-dependent vesicle fusion and acetylcholine release at nerve terminals.
The parent hexapeptide, Argireline, inhibited neurotransmitter release with a potency similar to botulinum toxin A but much lower efficacy; comparable mechanistic data for Snap-8 have not been published in indexed journals.
Reduced neurotransmitter release is proposed to lessen contraction of the facial muscles that form expression lines; this effect has not been isolated for Snap-8 in controlled human studies.

Human cosmetic study
PMID 39082657Clinical Safety and Efficacy Evaluation of a Dissolving Microneedle Patch Having Dual Anti-Wrinkle Effects With Safe and Long-Term Activities.
Shin JY, et al. · Ann Dermatol
In 24 healthy subjects, a microneedle patch containing acetyl octapeptide-3, ascorbyl glucoside and cyclic lysophosphatidic acid improved eye-wrinkle, hydration and elasticity measures versus a hyaluronic acid patch; the peptide's individual effect was not assessed.

Human cosmetic study
PMID 31134751Efficacy of bioactive peptides loaded on hyaluronic acid microneedle patches: A monocentric clinical study.
Avcil M, et al. · J Cosmet Dermatol
A 12-week single-center study of microneedle patches containing acetyl octapeptide-3 with other peptides, adenosine and seaweed extracts reported fewer fine lines and higher hydration and dermal density; the design did not isolate the peptide's effect.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
No peer-reviewed studies of Snap-8 alone are indexed; the two human studies tested multi-ingredient patches, were small, and did not report randomization, so the peptide's contribution is unknown. No adverse effects were reported in those studies, but long-term safety data are lacking. Snap-8 is a cosmetic ingredient, not an approved drug.
