5-Amino-1MQ
Also known as 5-amino-1-methylquinolinium, 5A1MQ, NNMTi
5-Amino-1MQ is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme linked to fat tissue metabolism and NAD+ levels. It has been studied in mouse models of obesity and muscle aging.

Overview
5-Amino-1MQ (5-amino-1-methylquinolinium) is a small molecule, not a peptide. It came from structure-activity studies of quinolinium compounds by researchers at the University of Texas Medical Branch, who designed the series to block NNMT, an enzyme overexpressed in white adipose tissue in obesity. In vitro, 5-amino-1MQ was membrane-permeable and did not inhibit related methyltransferases or NAD+ salvage enzymes. Rodent studies have examined obesity, liver fat, and aged skeletal muscle. No human trials have been reported in the indexed literature.
Class: Small-molecule nicotinamide N-methyltransferase (NNMT) inhibitor
Mechanism of action
How the compound acts, as described in the literature.
NNMT transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, producing 1-methylnicotinamide (1-MNA). This consumes SAM and diverts nicotinamide away from NAD+ salvage.
In adipocytes, 5-amino-1MQ and related inhibitors lowered intracellular 1-MNA, raised NAD+ and SAM, and suppressed lipogenesis.
In diet-induced obese mice, NNMT inhibition reduced body weight, white fat mass, and adipocyte size without reducing food intake. Later work reported improved glucose tolerance, insulin sensitivity, and liver fat.
In aged mice, NNMT inhibition activated senescent muscle stem cells and was associated with larger muscle fibers and greater contractile force after injury, along with shifts in NAD+/NADH redox state.

Rodent and in vitro study
PMID 29155147Selective and membrane-permeable small molecule inhibitors of nicotinamide N-methyltransferase reverse high fat diet-induced obesity in mice.
Neelakantan H, et al. · Biochem Pharmacol
Methylquinolinium NNMT inhibitors were permeable and selective in vitro and raised NAD+ and SAM in adipocytes. In diet-induced obese mice, 5-amino-1MQ reduced body weight, white fat mass, adipocyte size, and cholesterol without changing food intake.

PMID 30753815Small molecule nicotinamide N-methyltransferase inhibitor activates senescent muscle stem cells and improves regenerative capacity of aged skeletal muscle.
Neelakantan H, et al. · Biochem Pharmacol
In 24-month-old mice with induced muscle injury, the NNMT inhibitor 5-amino-1MQ increased muscle stem cell proliferation and fusion, roughly doubled myofiber cross-sectional area, and raised peak torque by about 70%.

Rodent study
PMID 39161060Nicotinamide N-methyltransferase inhibition mitigates obesity-related metabolic dysfunction.
Babula JJ, et al. · Diabetes Obes Metab
In diet-induced obese mice, 28 days of 5A1MQ dose-dependently limited weight and fat gain, improved glucose tolerance and insulin sensitivity, and reduced hepatic steatosis and liver enzymes. Pharmacokinetic and tissue distribution data were also reported.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
All published evidence comes from cell and rodent studies; no human safety, pharmacokinetic, or efficacy data are indexed on PubMed. Most studies come from a single group of investigators, including authors affiliated with the company Ridgeline Therapeutics, and have not been independently replicated. 5-Amino-1MQ is not an approved drug.
