Cardiogen
Also known as AEDR, Ala-Glu-Asp-Arg, H-Ala-Glu-Asp-Arg-OH
Cardiogen is a synthetic tetrapeptide, Ala-Glu-Asp-Arg, from the Khavinson series of short-peptide bioregulators. It has been studied in cell and rodent models related to heart tissue, cell proliferation, and apoptosis.

Overview
Cardiogen was developed at the St. Petersburg Institute of Bioregulation and Gerontology, the group led by Vladimir Khavinson, as part of a series of di-, tri-, and tetrapeptides intended to reproduce the activity of tissue-derived peptide extracts. The developers describe it as cardioprotective. Published work, largely from this group and often in Russian journals, reports effects on proliferation and apoptosis in rat myocardial tissue culture, on cytoskeletal protein expression in fibroblasts, and on transplanted tumor growth in rats. Independent replication is limited, and no controlled human trials are indexed in PubMed.
Class: Synthetic tetrapeptide (Khavinson short-peptide bioregulator)
Mechanism of action
How the compound acts, as described in the literature.
Proposed by its developers to act as a gene-regulating short peptide that reaches the nucleus and interacts with DNA or chromatin. This model has not been independently established.
In mouse embryonic fibroblasts, increased expression of the cytoskeletal proteins actin, tubulin, and vimentin and of the nuclear matrix proteins lamin A and lamin C.
In rat myocardial explants, stimulated cell proliferation and lowered p53 expression, which the authors interpreted as reduced apoptosis.
In rats with transplanted sarcoma, increased tumor-cell apoptosis and hemorrhagic necrosis, effects the authors attributed to action on tumor blood vessels.

2012
In vitro
PMID 22977870Tetrapeptide H-Ala-Glu-Asp-Arg-OH stimulates expression of cytoskeletal and nuclear matrix proteins.
Khavinson VKh, et al. · Bull Exp Biol Med
In cultured mouse embryonic fibroblasts, the tetrapeptide raised expression of actin, tubulin, and vimentin 2- to 5-fold and lamins A and C 2- to 3-fold. The authors linked this to its reported cardioprotective activity.
2009
Ex vivo tissue culture
PMID 20210190[The effect of the amino acids and cardiogen on the development of myocard tissue culture from young and old rats].
Chalisova NI, et al. · Adv Gerontol
In organotypic myocardial explants from 3- and 24-month-old rats, cardiogen stimulated cell proliferation at both ages, while few of 20 amino acids tested were active in old tissue. Cardiogen also reduced p53 expression. Article in Russian.
2009
Rodent study
PMID 20396706Tumor-modifying effect of cardiogen peptide on M-1 sarcoma in senescent rats.
Levdik NV, Knyazkin IV. · Bull Exp Biol Med
In senescent rats with transplanted M-1 sarcoma, cardiogen increased tumor-cell apoptosis and inhibited tumor growth in a dose-dependent way through hemorrhagic necrosis, without a direct cytostatic effect.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Evidence consists of a few small cell, tissue-culture, and rat studies, mostly from the developer's group and often in Russian-language journals, without independent replication. No controlled human trials or human safety data are indexed, and Cardiogen is not an approved drug.
