KPV
Also known as Lys-Pro-Val, α-MSH(11–13)
KPV is the tripeptide lysine-proline-valine, the C-terminal sequence of alpha-melanocyte-stimulating hormone. It is studied for anti-inflammatory effects, mainly in cell and mouse models of intestinal inflammation.

Overview
Alpha-melanocyte-stimulating hormone (α-MSH) is a 13-amino-acid peptide with anti-inflammatory activity, and work in the late 1980s traced part of that activity to its C-terminal tripeptide, Lys-Pro-Val. Research on KPV has focused on colitis models, where it is taken up by the intestinal peptide transporter PepT1 and reduces inflammatory signaling in epithelial and immune cells. Later work has examined delivery systems, such as hyaluronic acid-coated nanoparticles, designed to carry it to the inflamed colon. Published research is preclinical, and no human trials of KPV are indexed on PubMed.
Class: Melanocortin-derived anti-inflammatory tripeptide
Mechanism of action
How the compound acts, as described in the literature.
Transported into intestinal epithelial cells and immune cells by the proton-coupled peptide transporter PepT1, where nanomolar concentrations reduced cytokine-induced NF-κB and MAP kinase activation.
Lowered secretion of pro-inflammatory cytokines in cultured cells and reduced colonic inflammatory infiltrates and myeloperoxidase activity in mouse colitis models.
Its anti-inflammatory effects in mice appear at least partly independent of melanocortin-1 receptor signaling, since KPV remained active in mice lacking functional MC1R.
Hyaluronic acid-functionalized nanoparticles, which target CD44, have been used to deliver KPV to colonic epithelial cells and macrophages in mice.

Selected studies, each linked to its PubMed record. Summaries describe what the studies reported.
2008
In vitro and mouse study
PMID 18061177PepT1-mediated tripeptide KPV uptake reduces intestinal inflammation.
Dalmasso G, et al. · Gastroenterology
In intestinal epithelial and T-cell lines, nanomolar KPV reduced cytokine-induced NF-κB and MAP kinase signaling and cytokine secretion, dependent on PepT1 uptake; oral KPV reduced DSS- and TNBS-induced colitis in mice.
2008
Mouse study
PMID 18092346Melanocortin-derived tripeptide KPV has anti-inflammatory potential in murine models of inflammatory bowel disease.
Kannengiesser K, et al. · Inflamm Bowel Dis
KPV improved weight recovery and reduced histological inflammation and myeloperoxidase activity in DSS and T-cell transfer colitis; in MC1R-deficient mice it prevented DSS-related deaths, suggesting partial independence from MC1R signaling.
2017
Mouse study
PMID 28143741Orally Targeted Delivery of Tripeptide KPV via Hyaluronic Acid-Functionalized Nanoparticles Efficiently Alleviates Ulcerative Colitis.
Xiao B, et al. · Mol Ther
Hyaluronic acid-functionalized nanoparticles delivered KPV to colonic epithelial cells and macrophages in mice with colitis, with better mucosal protection and lower TNF-α than nanoparticles lacking the hyaluronic acid coating.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
All published evidence comes from cell and rodent studies, mainly colitis models, and no human trials are indexed, so human safety and pharmacokinetics are unknown. KPV is not an approved drug.
