Mazdutide
Also known as LY3305677, IBI362
Mazdutide is a peptide that activates both the GLP-1 and glucagon receptors. It is approved in China for chronic weight management and has been studied in obesity and type 2 diabetes.

Overview
Mazdutide was discovered by Eli Lilly as LY3305677 and is developed in China by Innovent Biologics as IBI362. Like survodutide, its design draws on oxyntomodulin, a gut hormone with dual glucagon and GLP-1 receptor activity, and it carries a fatty-acid side chain that extends its half-life. China's drug regulator approved it for chronic weight management in June 2025. The GLORY trials studied it in obesity and the DREAMS trials in type 2 diabetes, mostly in Chinese adults, with a phase 2 study in the United States.
Class: Dual glucagon and GLP-1 receptor agonist
Mechanism of action
How the compound acts, as described in the literature.
Activates the GLP-1 receptor, increasing glucose-dependent insulin secretion, slowing gastric emptying, and reducing appetite.
Activates the glucagon receptor, which in the liver promotes fatty acid oxidation and energy expenditure. This is proposed to add to weight and liver fat reductions beyond GLP-1 receptor agonism alone.
The GLP-1 component offsets the glucose-raising effect of glucagon receptor activation. In type 2 diabetes, a phase 3 trial reported larger HbA1c reductions than with dulaglutide.

2025
Phase 3 RCT
PMID 40421736Once-Weekly Mazdutide in Chinese Adults with Obesity or Overweight.
Ji L, et al. · N Engl J Med
GLORY-1 randomized 610 Chinese adults with overweight or obesity. At week 32, mean body-weight change was -10.09% and -12.55% at two dose levels versus +0.45% with placebo; reductions continued to week 48.
2026
Phase 3 RCT
PMID 42251595Treatment With 9-mg Mazdutide for Weight Reduction in Chinese Adults With Obesity: The GLORY-2 Randomized Clinical Trial.
Gao L, et al. · JAMA
GLORY-2 randomized 462 Chinese adults with a BMI of 30 or higher. At 60 weeks, mean body-weight change was -16.65% with mazdutide versus -1.50% with placebo.
2026
Phase 3 RCT
PMID 41407860Mazdutide versus dulaglutide in Chinese adults with type 2 diabetes.
Guo L, et al. · Nature
DREAMS-2 randomized 731 adults with type 2 diabetes on oral glucose-lowering drugs to mazdutide or dulaglutide for 28 weeks. Both mazdutide dose levels reduced HbA1c and body weight more than dulaglutide.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Mazdutide is approved only in China, and nearly all pivotal data come from Chinese adults, so results may not generalize to other populations; long-term outcome data are not yet available. In GLORY-2, vomiting (53.1% versus 1.3%), nausea (46.9% versus 3.2%), and diarrhea (39.4% versus 6.5%) were more frequent than with placebo, and 2.9% discontinued for adverse events versus 0%. DREAMS-2 also reported more gastrointestinal events than with dulaglutide.
