Overview
SLU-PP-332 is a synthetic small molecule, not a peptide; the SLU prefix refers to Saint Louis University, where the compound series originated. It targets the estrogen-related receptors (ERRs), nuclear receptors that do not bind estrogen despite their name. ERRs regulate genes for mitochondrial biogenesis, fatty acid oxidation, and oxidative muscle fibers, a program also switched on by endurance exercise. SLU-PP-332 activates all three ERR isoforms, with highest potency at ERRα. Published research consists of cell and mouse studies of exercise capacity, metabolic syndrome, heart failure, and kidney aging.
Class: Synthetic pan-estrogen-related receptor (ERRα/β/γ) agonist
Mechanism of action
How the compound acts, as described in the literature.
Acts as an agonist at ERRα, ERRβ, and ERRγ, with highest potency at ERRα. ERRs are constitutively active nuclear receptors that work with the coactivator PGC-1α to drive oxidative metabolism genes.
In muscle cells it increased mitochondrial function and respiration. In mice it increased type IIa oxidative fibers and induced an acute aerobic exercise gene program that depended on ERRα.
In obese mouse models it increased energy expenditure and fatty acid oxidation, decreased fat mass accumulation, and improved insulin sensitivity.
In aged mice it reversed kidney mitochondrial dysfunction and inflammation, with effects involving cGAS-STING and STAT3 signaling.

ERR levels were reduced in aging human and mouse kidneys. In 21-month-old mice, eight weeks of SLU-PP-332 reversed albuminuria, podocyte loss, mitochondrial dysfunction, and increases in inflammatory cytokines.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
All published evidence comes from cell and mouse studies; no human studies or formal toxicology reports are indexed on PubMed. The core exercise and metabolic studies come from one research group, and several authors declared stock holdings in Myonid Therapeutics, a company developing ERR-based therapeutics. SLU-PP-332 is not an approved drug.
