Overview
CJC-1295 with DAC combines a stabilized GRF (1-29) sequence with a reactive maleimide linker, the drug affinity complex (DAC), which bonds covalently to serum albumin after administration. Attached to albumin, the peptide escapes rapid clearance and continues to stimulate the pituitary GHRH receptor for days. It reached early clinical studies in healthy adults and was investigated as a long-acting alternative to daily GHRH, but it was not developed to approval.
Class: Long-acting GHRH analogue (albumin-binding)
Mechanism of action
How the compound acts, as described in the literature.
Acts as an agonist at the pituitary GHRH receptor, increasing cAMP and stimulating growth hormone release.
The DAC linker reacts with cysteine-34 of serum albumin, forming a bioconjugate that circulates far longer than unmodified peptide.
Prolonged receptor stimulation raises GH concentrations for several days and IGF-1 for over a week after a single administration in published studies.
Sustained exposure shifts the GH profile toward elevated baseline levels rather than discrete pulses alone.

Rodent study
PMID 15817669Human growth hormone-releasing factor (hGRF)1-29-albumin bioconjugates activate the GRF receptor on the anterior pituitary in rats: identification of CJC-1295 as a long-lasting GRF analog.
Jetté L, et al. · Endocrinology
In rats, albumin-bound hGRF(1-29) conjugates activated the pituitary GRF receptor; the work identified CJC-1295 as a long-lasting analogue.
2006
Randomized placebo-controlled trials
PMID 16352683Prolonged stimulation of growth hormone (GH) and insulin-like growth factor I secretion by CJC-1295, a long-acting analog of GH-releasing hormone, in healthy adults.
Teichman SL, et al. · J Clin Endocrinol Metab
In healthy adults, single doses raised mean GH 2- to 10-fold for 6 days or more and IGF-I 1.5- to 3-fold for 9–11 days; estimated half-life was 5.8–8.1 days. No serious adverse reactions were reported.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Human data come from short trials in small numbers of healthy adults; effects of sustained GH and IGF-1 elevation over longer periods were not studied. The compound was never approved, and clinical development did not continue beyond early-phase studies.
