Overview
Hexarelin is a synthetic hexapeptide related to GHRP-6, with a 2-methyl-tryptophan substitution that increases potency and stability. In human pharmacology studies it released growth hormone by several routes of administration. Work in the late 1990s showed that hexarelin also binds CD36, a scavenger receptor expressed in the heart, independently of the ghrelin receptor. This cardiac binding prompted research into its cardiovascular effects. It has not been approved for any indication.
Class: Ghrelin receptor agonist; CD36 ligand
Mechanism of action
How the compound acts, as described in the literature.
Agonist at the ghrelin receptor (GHS-R1a), stimulating strong GH release.
Also binds CD36, a scavenger receptor in cardiac tissue, a site distinct from the classic GH secretagogue receptor.
CD36 binding has been linked in animal studies to effects on coronary perfusion and cardiac function.
Can raise ACTH, cortisol and prolactin, like other non-selective GHRPs.
Published research
Selected studies, each linked to its PubMed record. Summaries describe what the studies reported.
1994
Human pharmacology study
PMID 8126144Growth hormone-releasing activity of hexarelin, a new synthetic hexapeptide, after intravenous, subcutaneous, intranasal, and oral administration in man.
Ghigo E, et al. · J Clin Endocrinol Metab
Showed that hexarelin released growth hormone in humans by several routes of administration, characterizing its potency as a GH secretagogue.
1999
Molecular / tissue study
PMID 10532947Identification and characterization of a new growth hormone-releasing peptide receptor in the heart.
Bodart V, et al. · Circ Res
Identified a cardiac binding site for hexarelin distinct from the pituitary GH secretagogue receptor, later characterized as CD36.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Human studies are small and short. Repeated exposure has been reported to blunt the GH response over time. Cardiovascular findings come mainly from animal and tissue studies. It is not approved for any use.
