IGF-1 LR3
Also known as Long R3 IGF-I, LR3-IGF-1

Overview
IGF-1 LR3 is a recombinant analogue of human IGF-I with an arginine in place of glutamate at position 3 and a 13-amino-acid N-terminal extension. These changes sharply reduce binding to IGF-binding proteins, which normally sequester IGF-I, so more of the analogue stays free to activate the IGF-1 receptor. It was developed in the early 1990s and is widely used in cell culture media. Infusion and injection studies in animals showed greater potency than native IGF-I.
Class: Recombinant IGF-I analogue
Mechanism of action
How the compound acts, as described in the literature.
Activates the type 1 IGF receptor (IGF-1R), a receptor tyrosine kinase that signals through the PI3K–Akt and MAPK pathways.
The Arg3 substitution and N-terminal extension greatly reduce binding to IGF-binding proteins, increasing the free, active fraction.
IGF-1R signaling promotes cell growth, protein synthesis and glucose uptake in target tissues.
Lower binding-protein affinity extends effective activity compared with native IGF-I in animal models.
Published research
Selected studies, each linked to its PubMed record. Summaries describe what the studies reported.
1992
In vitro study
PMID 1378742Novel recombinant fusion protein analogues of insulin-like growth factor (IGF)-I indicate the relative importance of IGF-binding protein and receptor binding for enhanced biological potency.
Francis GL, et al. · J Mol Endocrinol
Showed that IGF-I analogues with reduced IGF-binding protein binding, including the Long R3 form, had markedly enhanced biological potency in vitro.
1996
Rodent study
PMID 8708565Superior potency of infused IGF-I analogues which bind poorly to IGF-binding proteins is maintained when administered by injection.
Tomas FM, et al. · J Endocrinol
In rats, IGF-I analogues with weak binding-protein affinity were more potent than native IGF-I, both when infused and when injected.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
All published evidence is from cell culture and animal studies; there are no human trials. IGF-1 receptor signaling lowers blood glucose, and hypoglycemia is a recognized effect of IGF-I class compounds. Long-term effects on cell proliferation are not characterized.
