MGF
Also known as Mechano growth factor, IGF-1Ec E-peptide
A peptide based on the C-terminal E-domain of IGF-1Ec, a splice variant first identified in muscle subjected to stretch. Its biological role is disputed.

Overview
Mechano growth factor (MGF) refers to an IGF-1 splice variant, IGF-1Ec in humans, cloned from skeletal muscle exposed to stretch and damage. Research products usually consist of a synthetic peptide matching the variant's unique C-terminal E-domain. Early studies proposed that this E-peptide promotes myoblast proliferation separately from mature IGF-1. Later independent work by pharmaceutical laboratories could not reproduce those effects, and whether the E-peptide has a physiological function remains an open question.
Class: IGF-1 splice-variant E-peptide
Mechanism of action
How the compound acts, as described in the literature.
Derived from the C-terminal E-domain of the IGF-1Ec splice variant, which is upregulated in muscle after mechanical loading in animal studies.
Early work proposed that the E-peptide stimulates myoblast proliferation while mature IGF-1 drives differentiation.
The receptor or signaling pathway for the E-peptide itself has not been firmly established.
Independent replication studies reported no effect of the MGF peptide on myoblasts or muscle stem cells.

Animal / molecular study
PMID 8884603Cloning and characterization of an IGF-1 isoform expressed in skeletal muscle subjected to stretch.
Yang S, et al. · J Muscle Res Cell Motil
Cloned an IGF-1 isoform expressed in stretched skeletal muscle, the variant later termed mechano growth factor.
2002
In vitro study
PMID 12095637Different roles of the IGF-I Ec peptide (MGF) and mature IGF-I in myoblast proliferation and differentiation.
Yang SY, et al. · FEBS Lett
Proposed that the MGF E-peptide promotes myoblast proliferation while mature IGF-I promotes differentiation.
2014
In vitro replication study
PMID 24253050Mechano-growth factor peptide, the COOH terminus of unprocessed insulin-like growth factor 1, has no apparent effect on myoblasts or primary muscle stem cells.
Fornaro M, et al. · Am J Physiol Endocrinol Metab
Independent laboratories found that the MGF peptide, at concentrations up to 500 ng/ml, failed to increase proliferation of myoblasts or primary muscle stem cells, questioning its physiological role.
Evidence & safety
What the research does not show, and the adverse events and regulatory status it reports.
Key early findings were not reproduced by independent laboratories (Fornaro 2014), and the peptide's physiological role remains disputed. There are no human trials, and safety data are absent.
